چکیده
Epidemiological studies have shown that lung cancer is associated with tobacco and alcohol use, and is common in regions where these products are often consumed. Similar to other environmental toxins, tobacco requires metabolic activation and subsequent detoxification by a series of enzymes such as phase I enzymes i-e the cytochrome P450 enzymes (CYP) and phase II xenobiotic metabolizing enzymes i-e the glutathione S-transferases (GSTs). An association between CYP1A1 polymorphisms and lung cancer was first reported by Kawajri and co-workers in 1990 in an asian population study (Kawajiri et al., 1990). Cytochrome P4501A1 (CYP1A1) metabolizes a number of suspected procarcinogens, especially, polycyclic aromatic hydrocarbons (PAHs) into highly reactive intermediates. These compounds are capable to bind with DNA to form adducts, which, if not repaired, may initiate or propagate carcinogenesis. Although PAHs are

Susceptibility of lung cancer with polymorphisms of CYP1A1, GSTM1, GSTM3, GSTT1 and GSTP1 genotypes in the population of Inner Mongolia region.
BACKGROUND To study the relationship of susceptibility to lung cancer with the gene polymorphisms of CYP1A1, GSTM1, GSTM3, GSTT1, GSTP1 and smoking status in Han and Mongolian populations of Inner Mongolia, an autonomous region of China. MATERIALS AND METHODS PCR-RFLP, allele-specific and multiplex PCR were employed to identify the genotypes of CYP1A1, GSTM1, GSTM3, GSTT1 and GSTP1 in a case-...

Biotransformation enzymes are related with lung cancer that arises as a consequence of exposure to mutagenic agents. CYP1A1 gene codifies the phase I enzyme, aryl hydrocarbon hydroxilase, belonging to the Cytochrome P450 system, that plays a major role in the bioactivation of tobacco procarcinogenes, while glutathione-Stransferases genes, GSTM1, GSTT1 and GSTP1, codify conjugation enzymes assoc...
متن کاملAn international case-control study of glutathione transferase and functionally related polymorphisms and risk of primary adult brain tumors.
BACKGROUND Glutathione transferases (GST) detoxify environmental and endogenous compounds and levels of two polymorphic GST proteins, GSTM3 and GSTP1, are high in the brain. Previous studies of GSTM3 and GSTP1 polymorphisms and adult brain tumor risk have produced inconsistent results, whereas the GSTM3 -63 variant is newly identified and, therefore, has not yet been studied in this context. We...
متن کاملCYP1A1, CYP2E1, GSTM1, GSTT1, GSTP1, and TP53 polymorphisms: do they indicate susceptibility to chronic obstructive pulmonary disease and non-small-cell lung cancer?
Gene polymorphisms of phase I (CYP1A1 and CYP2E of cytochrome P,) and phase II (GSTM1, GSTT1 and GSTP1 of glutathione-S-transferase,) enzymes and the TP53 tumor suppressor gene were studied as markers in a sample of 262 Brazilians of European descent, the sample consisting of 97 patients with non-small-cell lung cancer (NSCLC), 75 patients with chronic obstructive pulmonary disease (COPD) and 9...
متن کاملGenetic Polymorphisms of Phase II Metabolic Enzymes and Lung Cancer Susceptibility in a Population of Central South China
A case-control study was conducted for analyzing the genetic polymorphisms of phase II metabolic enzymes in 97 patients with lung cancer and 197 healthy subjects from Han ethnic group of Hunan Province located in Central South China. The results showed that the frequencies of glutathione S-transferase (GST) M1-null (GSTM1-) or GSTT1-null (GSTT1-) genotype alone, or combined form of both in lung...
